基于婴幼儿食品安全的叶黄素含量测定方法研究

文章建立一种高效、准确、稳定的高效液相色谱法(HPLC),用于测定婴幼儿配方食品中叶黄素的含量,以满足婴幼儿食品的安全监管及质量控制需求。样品经氢氧化钾溶液室温皂化使结合态叶黄素游离,采用乙醚-正己烷-环己烷(40+40+20,v/v/v)混合溶剂萃取,经C30反相色谱柱分离,以甲醇-水和甲基叔丁基醚(MTBE)为流动相进行梯度洗脱,经紫外检测器在445 nm波长下检测。结果表明,叶黄素在0.05~2.00 μg/mL范围内线性关系良好,回归方程为 y=762.207 008x-1.277 042 9,相关系数R2=0.998 07。方法的定量限(LOQ)为10 μg/100 g。在3个不同加标水平下,平均回收率为92.5%~95.8%,相对标准偏差(SRSD)为1.34%~5.39%(n=6)。该方法前处理简便,有效消除了婴幼儿配方食品中复杂基质的干扰,分离度高,重现性好,准确度符合食品安全检测要求,可作为婴幼儿配方食品中叶黄素日常监督检测和质量控制的可靠方法。

Abstract:

The study establishes an efficient,accurate and stable high-performance liquid chromatography (HPLC) method for the determination of lutein in infant formula foods,so as to meet the needs of food safety supervision and quality control of infant foods.The samples were liberated by saponification with potassium hydroxide solution at room temperature,and then extracted with a mixed solvent of diethyl ether-n-hexane-cyclohexane (40+40+20,v/v/v).The separation was performed on a C30 reversed-phase chromatographic column with gradient elution using methanol-water and methyl tert-butyl ether (MTBE) as the mobile phases,and the detection was carried out with an ultraviolet detector at the wavelength of 445 nm.The results show that lutein shows a good linear relationship in the range of 0.05~2.00 μg/mL,with the regression equation of y=762.207 008x-1.277 042 9 and the correlation coefficient R2=0.998 07.The limit of quantitation (LOQ) of the method is 10 μg/100 g.At three different spiked levels,the average recoveries range from 92.5% to 95.8%,and the relative standard deviations (SRSD) are between 1.34% and 5.39% (n=6).This method is simple in sample pretreatment,which can effectively eliminate the interference from the complex matrix of infant formula foods.It has high resolution,and good reproducibility and its accuracy meets the requirements of food safety detection,and can be used as a reliable method for the routine supervision,detection and quality control of lutein in infant formula foods.
